Combination Vaccines of Fasciola gigantica Saposin-like Protein-2 and Leucine Aminopeptidase

Saposin-like protein-2 (SAP-2) and leucine aminopeptidase (LAP) are major proteins involved in the digestive process of Fasciola gigantica (Fg). Both SAP-2 LAP highly expressed F. gigantica; therefore, they could be vaccine candidates for fasciolosis. The aims this study (1) to observe tissue expression (FgSAP-2) (FgLAP) by indirect immunofluorescence technique under confocal microscopy (2) test potentials individual combined recombinant (r) FgSAP-2 rFgLAP against Imprinting Control Region (ICR) mice (n = 10 per group). By immunofluorescence-confocal microscopy, FgLAP were localized caecal epithelium but at different sites: appeared small granules that distributed middle lower parts cytoplasm epithelial cells, while as a line or zone apical cells. For testing, percent protection rFgSAP-2 vaccines was 80.7 81.4% when compared with aluminum hydroxide (alum) adjuvant unimmunized controls, respectively. levels IgG1 IgG2a sera significantly increased single combine vaccinated groups control groups. Vaccinated showed reduced liver damage This indicates had higher potential than vaccine. These results support further testing application infection farmed livestock animals.
